Gregor Hološka was born in Myjava. He graduated from the Academy of Performing Arts in Bratislava in 2000. From 2000 to 2014, he was a permanent member of DJP in Trnava. From 2014 to 2023, he was a freelance performer, guest starring in several theatres — Arena, DPOH — and in several private productions. He has acted in television, film, radio, and dubbing.
He became a permanent member of the Slovak National Theatre in March 2023. He has been awarded the Literary Fund Prize for his theatre performances several times, e.g., in 2000, for the character of Charles Bovary and in 2003, for the character of Ďuro Ľavko in the production of Statky-zmätky. He received the annual Literary Fund Award for 2012 for his father's character in the production of The Builders of the Future. He received the 2019 Sun in the Net Film Award for Best Performance by a male actor in a supporting role for his portrayal of Gujda in Amnesty.
